Kuhner shakers are suitable for the cultivation of microbial, plant, human and animal cell cultures. In the registers below you will find more information about the different application areas and suitable product recommendations for optimal cultivation.

Stem cells

Stem cell cultivations run for several days or weeks and normally requires specific cultivation conditions, such as a low oxygen concentration (1-8%), high carbon dioxide concentration (4-10%), and a high relative humidity (>70%). Stem cells are mostly adherent cultures and are very sensitive to shear stress. Orbital shaken bioreactors are ideally suited for stem cell cultivations since they have a low local power input, therefore, significantly reducing the mechanical shear stress on the cultivated stem cells.

Cell culture

Cultivations of animal or human cell cultures run for several days or a few weeks. Usually, for cultivations they require a high carbon dioxide concentration (4-10%) and a high relative humidity (>70%). Cell cultures grow adherent or in suspension. Moreover, temperature shifts from 37 °C to 32 °C are often conducted during cultivations. When cultivating at a high relative humidity, this requires active dehumidifying of the incubator air to prevent condensation at the door or walls of the incubator shaker.

  • EPFL-table: This table accepts up to five (different) holders. To shake microtiter plates on the EPFL-table, a Duetz cover clamp is used which can accept up to 4 MTPs. Each holder has its own clamp handle allowing a complete Duetz cover clamp with 4 MTPS to be easily taken out of the incubator shaker and put on a clean bench while the other MTPs on the EPFL-table keep on shaking. 

  • Automatization: There are two main features Kuhner provides to support the automation. Optionally, an automatic door (ISF1-Z/X), which is activated by pressing a button on the shaker incubator or by using a foot switch. The other feature is a fixed stop position of the shaker table (not for 3 mm shaking diameter) which is important especially for a liquid handling robot.

  • For small shaking diameters (e.g. 3 mm) and high shaking frequencies (~1000 rpm): Duetz system for 96 well microtiter plates.

Cultivations in tubes

  • Shaker incubator with humidity and CO2 control (for larger flasks a humidity control is less significant since the influence of the evaporation rate decreases for larger filling volumes): ISF1-ZLT-XISF4-X.

Accessories:

  • Tray with pins : The tray allows parallel shaking of 7 x 3 L Corning Fernbach culture flasks, 7 x 5 L Corning flasks, or 7 x 5 L Thomson Optimum Growth flasks and is therefore ideally suited for cell cultures. Flasks are held in position by pins and are easily placed on the tray and removed just as quickly.

Yeasts & Fungi

Cultivations of yeasts and fungi run for approximately 3 to 5 days. Yeasts and fungi have a higher oxygen demand than animal or human cell cultures. A higher oxygen transfer can be achieved by decreasing the filling volume and increasing the shaking frequency. Moreover, active humidity control might be helpful to reduce evaporation losses. Filamentous fungi can increase the viscosity of the cultivation medium. In that case, large shaking diameters (50 or even 70 mm) should be chosen to prevent the occurrence of the “out-of-phase” phenomenon.

Bacteria

Cultivations of bacteria run for several hours or days. Bacteria usually have a much higher oxygen demand than animal or human cell cultures and often also a higher oxygen demand than yeasts or fungi. A high oxygen transfer can be achieved by small filling volumes and high shaking frequencies (300-350 rpm). Moreover, a humidity control is helpful to prevent high evaporation losses (especially for very small filling volumes and cultivation temperatures of 30-37 °C). For very small filling volumes, large shaking diameters (50 or even 70 mm) should be chosen to prevent the occurrence of the “out-of-phase” phenomenon.

Plant cells & algae

Cultivations of plant cell cultures and algae run for approximately 5 to 7 days. The oxygen demand lies in between animal cell cultures and bacteria. For longer cultivations, active humidity control is useful to decrease evaporation from the cultivation vessels. Many cultures require LED light for their photosynthesis reaction. Also, depending on the plant cell culture,  CO2 control might be necessary.

In many cases, different types of liquids must be thawed gently and evenly in a laboratory besides the cultivation of cell cultures and microorganisms. These liquids include for instance cultivation media, cell cultures, protein and enzyme solutions.

Mixing

In many cases, different kinds of liquids must be mixed or shaken in a laboratory besides the cultivation of cell cultures and microorganisms.   • EPFL-table: This table accepts up to five tube holders, each with a capacity of 24 x 50ml tubes. Each tube holder has its own clamp handle allowing it to be easily taken out of the incubator shaker and put on a clean bench while the other tubes keep on shaking.

  • Trays for centrifuge tubes: special trays which are available in two different sizes; with a capacity of 72 (3x24) 50 ml centrifuge tubes or 120 (5 x 24) 50 ml tubes.

  • 2D-Bag-tray (Cryo): tray for shaking (orbital or linear) 2D-Bags. Rubber bands secure smaller blood and cryo bags. It accepts various brands and sizes of pillow shaped single-use bags: 1 x 20 L (10 L working volume), 2 x 10 L (5 L working volume), 10 x 500 ml, or 12 x 250 ml.

  • 2D-Bag-tray (Flex): tray for shaking (orbital or linear) different types of 2D-bags. The tray consists of an aluminum base with a flexible mounting system on top. The mounting system can be manually adjusted to facilitate the controlled fixing of different sized cultivation bags. The mounting bars on both longitudinal sides can be moved up and down to hold the cultivation bag in place, while rubber bands will secure the smaller blood and cryo bags. The tray accepts various brands and sizes of pillow shaped single-use bags: 1 x 20 L (10 L working volume), 2 x 10 L (5 L working volume), 10 x 500 ml or 12 x 250 ml.

  • Incubator shaker for thawing with a temperature profile: ISF1-ZLT-XISF4-X.

Accessories:

  • Tray with support bars: This tray offers an alternative to both clamps and sticky mats and are ideal for tall containers. The tray is covered with a rubber mat and has an upright at each corner. E and EX size trays are fitted with four cross supports while the F tray is equipped with six. These support bars support and securely clamp the containers.
  • Trays for separating vessels/funnels: special tray for shaking separating vessels. This very flexible tray can take vessels of various sizes. It is also possible to add a second layer and double the capacity. Movable retaining rods allow vessels to be installed easily.
