Description

The Duetz-System is an integrated, easy-to-use, and cost-effective technology platform for the rapid handling and growth of large numbers of microbial strains, clonal libraries, and mutant banks in 24- or 96-well microtiter plates.

The system enables the growth in microplates of thousands of strains simultaneously without excessive repetitive handling: all handling is performed in parallel for sets of 24 or 96 strains. No robotic equipment is required. The system was originally developed and validated at the Institute of Biotechnology of the ETH Zurich.

Explanation of the most important distinguishing features and properties

Sandwich covers for cultures with high oxygen demand

Covers designed for cultures with high oxygen demand, such as fast-growing organisms like E. coli or yeast. The sandwich covers feature hole diameters optimized to support their increased oxygen requirements, ensuring enhanced gas exchange while minimizing evaporation during shorter cultivation periods of 1–4 days.

Sandwich covers for cultures with low oxygen demand

Covers designed for cultures with low oxygen demand, such as slow-growing organisms like mammalian or plant cells. The sandwich covers feature hole diameters optimized to meet their low oxygen needs, ensuring sufficient gas exchange while minimizing evaporation during longer cultivation periods of more than 4 days.

Sealing material: Soft (white) silicone

Soft (white and cellular) silicone is used as sealing material between sandwich cover and well plate for most applications and plates. It is soft and easily compressible, allowing it to seal wells even with low applied forces. Please note that this type of silicone does not absorb water: it features closed, air-filled cells and a solid, pore-free surface.

Sealing material: Solid (black) silicone

Solid (black) silicone is used as sealing material between sandwich cover and well plate for applications where minimizing silicone gas permeability is critical. Due to the stiffness of the material, more force is necessary to seal the wells gas tight. Solid silicone is also preferred for certain types of fungi that might otherwise grow into soft silicone materials.

Spacer pins

Spacer pins are integrated into the top of the cover to allow practical stacking of multiple plates with covers. They create an air gap between the plates, ensuring even and consistent airflow to all wells.

Side pins

Side pins made of metal are integrated into the inside of the cover to securely fix the layered system in place. Please note that covers with side pins are not compatible with all microtiter plates — the plates must have corresponding recesses at these positions.
